Effect of Hyung Bong Jeon Hwan on human female tumor cells and mouse immune cells

Abstract
The purpose of this research was to investigate effect of Hyung Bong Jeon Hwan(HBJH) on human female tumor cells and immune cells. The effects of HBJH on the proliferation of human female tumor cells, Balb/c 3T3 cells, mouse thymus cells and spleen cells were estimated by MTT colorimetric assay. DNA fragmentation was determined with flow cytometer after propidium iodide staining, T-lymphocyte subpopulation assay are carried out by cytofluorometric analysis after staining the cells with PWFITC-conjugated mAbs, and mitochondrial transmembrane potential was determined with flowcytometer after staining with DiOC_6. HBJH inhibited the proliferation of HeLa and MCF-7 cells, and did not affect the proliferation of Balb/c 3T3 cells. But HBJH inhibited the proliferation of thymus and spleen cells at 100 §¶/ml. HBJH induced apoptosis of T-lymphocytes, and increased CDW(Tc/Ts) cells in T-lymphocytes. HBJH stimulated nitric oxide production from mouse peritoneal macrophages in vivo. HBJH decreased lucigenin chemiluminescence, but incresed phagocytic activity of mouse peritoneal macrophages. HBJH induced apoptosis and reduced mitochondrial transmembrane potential of transplanted-L1210 cells in mice.
These results suggest that the anti-tumor activity of HBJH was induced by cytotoxicity on tumor cells, NO production and phagocytic activity of macrophages.
